Glycoside from Butea Superba Roxb and their cASVIP Phosphodiesterase
Inhibitory Activity
Abstract
Flavonoid(3,7,3'-trihydroy-4'-methoxyflavaone)(1)
and flavonoid glycosides dihydoxy-4'-methuxyflavone-7-0-p.-D-glucopyranoside)
(2) were isolated from the roots of Butea Superba Roxb. The structures were
determined on the basis of spectral analysis 2d-nmr lechiik-lues. These
compounds show strong inhibitory effects on cAMD phospodiesterase.
Introduction
Butea Superba Roxb. is a herb in the Family
Papilionaceae and has the characteristics of a crawer that wraps itself around
large trees. One branch has 3 leaves, the flowers are a yellowish color and
the plant grow's out in the open. The long roots of the plant are buried
under the "the round like roots of the yam." This type of plant
reproduces through seeds and the separation of its roots. This plant
can be found growing in forests in the northern regions, in the eastern
regions and along Karnchanaburi Province. The roots and stem of the plant are used for
medicines for strength and power. In addition, the roots and stem of the
plant are considered to help increase male sexual performance. Thus, this
plant has come to be known as one type of miracle herb. The bioactivity of
each constituent was tested, especially the cAMP phophodiesterase inhibitory
effects in controlling a number of diseases(1). Therefore, the research
conducted was designed to test the hypothesis that the chemical constituents
of this plant is actually capable in enhancing certain physiological
functions.
Results
From testing the bioactivity of
Flavonoid and Flavonoid glycoside, it was found that both of these compounds
were effective in inhibiting cAMP phosphodieterase. The result of inhibiting
cAMP phosphodiesterase of both these compounds was greater than theophylline
(IC50 =615 u.g/mL) and caffeine (IC50 = 420 u.g/rnL). The cAMP
phosphodiesterase enzyme has a main function in the hydrolysis of the
intracellular cAMP. Some compounds that
inhibit cAMP phosphodiesterase are therefore capable of stimulating the
functioning of the central nervous system (CNS) and stimulating the
functioning of cells.
Butea superba Roxb-
has a complex dynamic and has been shown effective in inhibiting the cAMP
phosphodieslerase enzyme. The
attributes of this herb in increasing male sexual performance is most
probably due to inhibition of cAMP phosphodiesterase, elevations of
androsteinedione and testosterone. Studies have demonstrated
that Butea Superba increases LuteInizing hormone, and
Follicle stimulating hormone. It has been
demonstrated in these same studies an androgenic effect as well as an
anti-estrogenic effect. Once could speculate Butea Superba could
help support heart health because of it's vasodilation properties.
Phosphodiesterase inhibitors like caffeine have been linked with lower
incidence of type 2 diabetes.
In a joint effort between Thai scientist and
scientists in the Netherlands the following anti-oxidant fractions have been
identified in Butea Superba:
procyanidin B2, (-)-epicatechin and procyanidin B5. Butea Superba also
contains the sterol compounds
